Wanneer de kwaliteit van een drugszending ernstig wordt aangetast worden drugssmokkelaars Man (Laurence Fishburne) en The Cook (Nicolas Cage) er door The Boss (Barry Pepper) op uitgestuurd om de supply chain van het drugscartel na te gaan. Ze volgen de cocaïne van The Farmer (Clifton Collings Jr.) zijn boerderij door de jungles van Colombia, via bergen, douane, gangsters en koeriers naar Canada. Een gevaarlijk avontuur met een hoog tempo terwijl The Agent In Charge (Leslie Dibb) ze ook nog eens op de hielen zit.
