Creation is gebaseerd op het boek Annie's Box van Kandal Keynes over de relatie tussen Charles Darwin en zijn tien jaar oude dochter Annie, wiens vroege dood Darwin diep beïnvloedde in zijn wantrouwen jegens God.
De film toont hoe Darwin een balans probeert te vinden tussen zijn revolutionaire theorieën over evolutie en de relatie met zijn zwaar gelovige vrouw. Als zijn dochter overlijdt, raakt hij lichamelijk en geestelijk in een neerwaartse spiraal. Haar geest is het echter die hem uit die donkere tijd redt. Dan is hij ook in staat het opzienbarende boek te schrijven dat de wereld zou veranderen: The Origin of Species.
