Als Supermans Cousine balanciert Kara Danvers (Supergirl) ihre Arbeit als Reporterin für CatCo Worldwide Media und für das Department of Extra-Normal Operations (DEO), eine streng geheime Regierungsorganisation, deren Mission es ist, National City - und die Erde - vor unheimlichen Bedrohungen zu beschützen. Bei der DEO arbeitet Kara für J'onn J'onzz (David Harewood), den Martian Manhunter, und mit ihrer Schwester Alex und ihrem besten Freund Winn Schott (Jeremy Jordan) zusammen. Und in Karas Leben finden wir auch James Olsen (Mehcad Brooks), einen Fotojournalist, der u. a. als Guardian, ein maskierter Vigilante, tätig ist, Lena Luthor (Katie McGrath) und Mon-El von Daxam (Chris Wood), dessen Planet von Kryptons Zerstörung verwüstet wurde. In der dritten Staffel müssen Kara und das Team gegen eine neue Bedrohung in Form von DC Worldkiller Reign kämpfen.
