Als Paul Castellano, Führungsmitglied der Cosa Nostra, 1986 auf offener Straße in Manhattan erschossen wird, rückt Gotti, der den Mord skrupellos selbst beauftragt hat, zum Mafiaboss von New York auf. Während Ehefrau Victoria (Kelly Preston) ihm den Rückhalt in einem scheinbar bürgerlichen Leben gibt, scheint Gotti lange Zeit unantastbar zu sein, weswegen er auch den Spitznamen Teflon-Don erhält, da alle Versuche der Strafverfolgung ihn einzusperren, spurlos an ihm abperlen. Doch allmählich gerät Gottis Imperium ins Wanken und auch sein Sohn John Gotti Jr. (Spencer LoFranco) fasst einen verhängnisvollen Entschluss.
